The (European) Protein Homeostasis Network
Our bodies rely on a sophisticated internal system, often called the protein homeostasis network, to keep our cells functioning correctly. This network is responsible for every step of a protein’s life, from its initial creation (protein synthesis) to ensuring it folds into the right three-dimensional shape (protein folding) to perform its job. It also handles the transport of proteins to their correct locations (protein trafficking) and the efficient removal of damaged or unneeded proteins (protein degradation) to prevent harmful buildup.
Recent research from across Europe has significantly advanced our understanding of this vital network. Scientists are exploring the fundamental ways cells manage their proteins, including how they regulate protein production and how specialized helper proteins, called molecular chaperones, assist in proper folding. They are also investigating key degradation pathways, such as the ubiquitin-dependent system, where proteins are tagged for destruction by cellular machinery called proteasomes.
A major focus is on how this network operates within specific cell compartments, like the endoplasmic reticulum and mitochondria, which are crucial for protein processing and energy production, respectively. Disruptions in protein management within these organelles are linked to various health issues, including aging, inflammation, and neurodegenerative diseases like Alzheimer’s and Parkinson’s. This collaborative European effort is not only deepening our knowledge of these complex cellular processes but also paving the way for new therapeutic strategies to combat diseases caused by protein imbalances.
